Kato, Laszlo (Institut de Microbiologie et d'Hygiène de l'Université de Montréal, Montreal, Quebec, Canada), and Bela Gozsy. Limited multiplication of Mycobacterium lepraemurium in parabiotic culture, as influenced by osmolarity of an alkaline-galactomannan medium. J. Bacteriol. 91:1859-1862. 1966.-Limited multiplication of Mycobacterium lepraemurium has been achieved in an alkaline (pH 8.4) galactomannan-containing medium, when cultivated alone or in parabiosis with a feeder strain, Torula minuta. Hyperosmolarity (NaCl, 2.0%) enhanced multiplication in both cases. With 2.0% NaCl in the medium, selective lysis of the feeder cells occurred, without damage to M. lepraemurium. Multiplication depends more on the physical properties (viscosity, hyperosmolarity, and alkaline pH) of the medium than on its chemical composition. The described conditions are proposed as a model for cultivation trials with other "apparently noncultivable" microorganisms.

Quantitative multiplication of Mycobacterium lepraemurium in a cell-free liquid medium (NC-5).
Mycobacterium Zepraemurium multiplies in a cell-free liquid medium, referred to as NC-5, which is enriched Kirchner medium plus goat serum, a-ketoglutaric acid, cytochrome c, haemin, and L-cysteine. At 30 "C, the bacilli gradually elongated before multiplying 100to Iooo-fold. The maximum number of bacilli is reached after 8 weeks' incubation. The generation time of Mycobacterium lepraemurium is...

Inability to cultivate Mycobacterium leprae in vitro has been regarded as the bottleneck which obstructs research on modes of transmission, pathogenesis, immunization and the treatment of leprosy . The bottleneck in the cultivation problem always has been that of gaining significant information from an organism that has given no useful response to intensive inquiries by the disciplines of bioch...
متن کاملRadiometric measurement of metabolic activity of Mycobacterium lepraemurium.
A sensitive and nondestructive radiometric method has been applied to the detection of metabolism of Mycobacterium lepraemurium, as a model for the study of the metabolism and substrate requirements of M. leprae. The method is based on the measurement of the (14)CO(2) produced through the bacterial conversion of [U-(14)C]acetate or [U-(14)C]glycerol by 7 x 10(9) bacteria suspended in 10 ml of e...
متن کاملEnhancement of growth of Mycobacterium lepraemurium in macrophages by gamma interferon.
Gamma interferon, an immune lymphokine that protects mouse macrophages against infection by several parasites, was ineffective against Mycobacterium lepraemurium. On the contrary, it significantly stimulated multiplication of M. lepraemurium in the macrophages. Simultaneous treatment of macrophages with gamma interferon and interleukin-4 or interleukin-2 or a combination of all three did not en...
متن کاملGrowth of Mycobacterium lepraemurium in cultures of macrophages obtained from various sources.
Studies were made on the growth of Mycobacterium lepraemurium in cultures of macrophages obtained from various sources, such as bone marrow, spleen, and blood of mice. Macrophages were maintained in good condition for more than 12 weeks. Marked intracellular multiplication of M. lepraemurium was observed in cultures from all three sources. Whereas M. lepraemurium freshly prepared from the anima...
